Researchers identify mechanism that makes breast cancer invasive

A new study has identified a key mechanism that causes breast cancer to spread. The research, published by Cell Press on March 30th in the journal Molecular Cell, enhances our knowledge about the signals that drive cancer metastasis and identifies new therapeutic targets for a lethal form of invasive breast cancer that is notoriously resistant to treatment.

Amplification of the gene for ErbB2 has been linked with aggressive forms of breast cancer and is associated with a poor outcome. Although ErbB2-positive breast cancers are routinely treated with chemotherapy and the ErbB2-targeted drug trastuzumab (Herceptin), this treatment often fails. "Nearly half of breast cancers with high ErbB2 expression are either initially non-responsive or eventually develop resistance to trastuzumab," explains senior study author, Dr. Tuula Kallunki, from the Danish Cancer Society Research Center. "Therefore, a better understanding of the molecular basis of ErbB2-induced malignancy is critical for further development of targeted treatments."

In the current study, Dr. Kallunki and colleagues discovered that ErbB2 induces expression of proteins called cathepsins. Previous work has linked cathepsins with metastasis of pancreatic cancer, specifically with tumor cell invasion of surrounding tissues and with the establishment of tumor blood supply. Using a three-dimensional model system to study ErbB2-induced cellular invasion, the researchers demonstrated that inhibition of cathepsins prevented ErbB2-driven breast cancer invasion. They went on to identify components of the complex signaling network linking ErbB2 and cathepsin expression with cancer invasiveness.

Taken together, the results demonstrate that the invasive behavior of ErbB2-positive cancer cells is dependent on cathepsin activity and uncover a molecular link between ErbB2 and breast cancer invasion. "Our work has introduced a cellular model system to study ErbB2-induced invasion and identified an invasion-promoting signaling network," concludes Dr. Kallunki. "In addition, our findings provide several potential therapeutic targets against ErbB2-driven invasive cancers."

Agreeing a deal, whether it?s a sale or purchase, partnership or hire, is relatively easy in principle. We each have something the other wants and there?s a deal to be done.

Negotiating the detail, however, is less simple. Here are some tips that we think can be applied across a huge variety of negotiation scenarios. Some of them may seem obvious, but they can be easily lost in the heat of the moment. Pick out the ones that you may forget, and revisit them in the days before you know a crucial piece of negotiation is looming.

  • Get rid of the perception that negotiating is about two Rottweilers locked in a room, with one eventually having to roll over. In Roger Fisher and William Ury?s negotiating text, Getting to Yes, the experts suggest that instead of viewing your counterpart as the adversary, you focus instead on the merits of the case and search for ways to find common ground. ?Attack? the underlying issue, rather than the other negotiator.
  • Have a goal in mind at the start of the discussion, for example, a figure you are happy to pay or accept for the provision of your goods or services. You will never finish negotiations for ?the best deal? if you dont know what it is.
  • Do your research. Find out as much as you can in advance about the other party and assume they will have done the same. Look on their website, read their LinkedIn profile, speak to contacts in your network who know them and how they operate
  • Spend time listening: the other person wants to deal as well and it?s important to understand their position. The 30/70 rule works well. Ask open-ended questions that won?t just get yes/no answers and listen for 70% of the time.
  • Don?t use the word ?between?. It often feels reasonable?and therefore like progress?to throw out a range. Saying ?I can do this for between ?10,000 and ?15,000? is tantamount to a concession, and any shrewd negotiator with whom you deal will swiftly hone in on the cheaper price or the later deadline.
  • Intimate that there is someone else you must speak to prior to saying yes, perhaps an investor, a partner, or a member of your advisory board. While you will almost certainly be making the decision yourself, you do not want the opposing negotiators to know that you are the final decision maker, just in case you get cornered as the conversation develops. You?ll almost always benefit from an extra 24 hours to consider the offer.
  • Once you make an offer, wait for a response before making another offer. By waiting, you avoid the possibility of rejecting your own offer and making further concessions in a revised offer. Remember the 30/70 rule.

Building your own website can drive you to drinkThe real estate brokerage community is a big proponent of having a professional do a job that a person is not trained to do. It is my view that we more than bring our value to a transaction by brokering the largest piece of business most people will ever undertake. We provide a marketing machine and exposure that a DIY lay person could only dream of. We protect them from themselves in negotiations.?We share our prodigious knowledge and contacts by pointing clients in the direction of the best lawyer, CPA, loan officer, contractor or tradesperson. With very rare exception, I truly believe that you have to be an idiot to not use a broker in a real estate transaction. My colleagues prove me right every day.?

Then, to save $500, real estate agents all over the country lose time, sleep and sanity building their own website instead of getting a professional to do it for them. Like some for sale by owners, a few agents do succeed and gain a valuable skill. They can build a website. I have probably built 20 websites myself. I can tell you from personal experience that consumers are far more concerned with the content I create than who did the code behind the scenes. If you are one of those agents who has successfully built an awesome website and know the ins and outs of Wordpress, good on ya. Much props. Way to go. Atta girl. Atta boy. You saved $500. You saved $1000.?

Wait, no you didn't. You saved writing some checks, but your time is worth something. Time that you could have spent selling real estate, getting another listing, or putting a deal together was spent learning CSS code or installing a plugin. Or, you could have spent the time sleeping. Or relaxing (remember that?). Or creating content. You broke even. You may have even lost money.?

I am not advocating that real estate agents not be tech-savvy. We should be cutting edge. But we should ?master technology to help us sell real estate better and in greater volume. Anything far beyond that is either?

Therapeutic; or
The Booby Prize.

If you dig building websites on your own and savor the challenge and all that good stuff, then by all means don't stop. You are getting something more from your efforts than a website. You are relaxing. It is your model train set. It is golf. I get that.

But if it isn't fun anymore, or the results are disappointing, you need to get help. Think of it as deciding that it is time to list with a broker now that the FSBO thing didn't work out. I don't mow my own lawn. I don't have time. I don't change my own oil. And this past February, I decided to hire an expert to build my Wordpress site from the ground up. I paid them $699 and I got a site I am really happy about with IDX, Altos Research, all my blog content imported over, and a bunch of other nice things. Do I still need to learn a ton of stuff? Of course- and the company does webinars several times a week. But am I going to be one of those people in Facebook support groups asking for help in fixing my site because I crashed it trying to do something exotic on my own? Not a chance. I'll spend my time creating content and selling real estate.?

The idea that you need to build your own Wordpress site to run with the big dogs may be costing you money. Hire a pro and devote your valuable time and focus to selling real estate.?

Supercomputing the difference between matter and antimatter

ScienceDaily (Mar. 29, 2012) ? An international collaboration of scientists has reported a landmark calculation of the decay process of a kaon into two pions, using breakthrough techniques on some of the world's fastest supercomputers. This is the same subatomic particle decay explored in a 1964 Nobel Prize-winning experiment performed at the U.S. Department of Energy's Brookhaven National Laboratory (BNL), which revealed the first experimental evidence of charge-parity (CP) violation -- a lack of symmetry between particles and their corresponding antiparticles that may hold the answer to the question "Why are we made of matter and not antimatter?"

The new research -- reported online in Physical Review Letters March 30, 2012 -- helps nail down the exact process of kaon decay, and is also inspiring the development of a new generation of supercomputers that will allow the next step in this research.

"The present calculation is a major step forward in a new kind of stringent checking of the Standard Model of particle physics -- the theory that describes the fundamental particles of matter and their interactions -- and how it relates to the problem of matter/antimatter asymmetry, one of the most profound questions in science today," said Taku Izubuchi of the RIKEN BNL Research Center and BNL, one of the members of the research team publishing the new findings. "When the universe began, did it start with more particles than antiparticles, or did it begin in a symmetrical way, with equal numbers of particles and antiparticles that, through CP violation or a similar mechanism, ended up with more matter than antimatter?"

Either way, the universe today is composed almost exclusively of matter with virtually no antimatter to be found.

Scientists seeking to understand this asymmetry frequently look for subtle violations in predictions of processes described by the Standard Model. One property of these processes, CP symmetry, can be explored by comparing two particle decays -- the decay of a particle observed directly and the decay of its anti-particle, viewed in mirror reflection. "C" refers to the exchange of a particle and its antiparticle (which is exactly the same but with opposite charge). "P" specifies the mirror reflection of this decay. But as the Nobel Prize-winning experiments showed, the two decays are not always symmetrical: In some cases you end up with extra particles (matter) and CP symmetry is "violated."

Exploring the precise details of the kaon decay process could help elucidate how and why this happens.

Supercomputing the decay process

The new calculation of one aspect of this decay, which required creating unique new computer techniques to use on some of the world's fastest supercomputers, was carried out by physicists from Brookhaven National Laboratory, Columbia University, the University of Connecticut, the University of Edinburgh, the Max-Planck-Institut f?r Physik, the RIKEN BNL Research Center (RBRC), the University of Southampton, and Washington University. The calculation builds upon extensive theoretical studies done since the first 1964 experiment and much more recent experiments done at CERN, the European particle physics laboratory, and at Fermi National Accelerator Laboratory.

The unprecedented accuracy of the measured experimental values -- which incorporate distances as minute as one thousandth of a femtometer (one femtometer is 1/1,000,000,000,000,000th of a meter, the size of the nucleus of a hydrogen atom) -- allowed the collaboration to follow the process in extreme detail: the decay of individual quarks (the subatomic components of many Standard Model particles) and the flitting in and out of existence of other subatomic particles. Viewing the picture from farther away -- a few tenths of a femtometer -- this basic process is obscured by a sea of quark-antiquark pairs and a cloud of the gluons that hold them together. At this distance, the gluons begin to bind the quarks into the observed particles. The last part of the problem is to show the behavior of the quarks as they orbit each other, moving at nearly the speed of light through a swarm formed from gluons and further pairs of quarks and antiquarks, and at last forming the pions of the decay under study.

To "translate" the mathematics needed to describe these interactions into a computational problem required the creation of powerful numerical methods and advances in technology that made possible the present generation of massively parallel supercomputers with peak computational speeds of hundreds of teraflops. (A teraflop computer can perform one million million operations per second).

The actual kaon decay described by the calculation spans distance scales of nearly 18 orders of magnitude, from the shortest distances of one thousandth of a femtometer -- far below the size of an atom, within which one type of quark decays into another -- to the everyday scale of meters over which the decay is observed in the lab. This range is similar to a comparison of the size of a single bacterium and the size of our entire solar system.

The collaboration carried out the computation using the methods of lattice quantum chromodyamics (QCD -- the theory that describes fundamental quark-gluon interactions), in which the decay is "imagined" as taking place within a lattice or grid of space-time points that can be entered into a computer. Then, the quantum fluctuations of the decay are calculated by a statistical method called the "Monte Carlo" method, which provides the most likely of the fluctuations as a result. The calculation required 54 million processor hours on the IBM BlueGene/P supercomputer installed at the Argonne Leadership Class Facility (ALCF) at Argonne National Laboratory. Earlier calculations were also done on Brookhaven's QCDOC (for QCD on a chip) supercomputer, a prototype for IBM's BlueGene series.

This calculation, when compared with predictions from the Standard Model, allows the scientists to determine another remaining unknown quantity important to understanding kaon decay and its relation to CP violation. A direct calculation of this remaining unknown quantity and a higher precision recalculation of the present result will be the focus of future research, requiring even more computing power.

"Fortunately," says co-author Peter Boyle of the University of Edinburgh, "the next generation of IBM supercomputers is being installed over the next few months in many research centers around the world, including the ALCF, the University of Edinburgh, the KEK laboratory in Japan, Brookhaven Lab, and the RBRC."

These new IBM BlueGene/Q machines are expected to have 10 to 20 times the performance of the current machines, Boyle explained. "With this dramatic boost in computing power we can get a more accurate version of the present calculation, and other important details will come within reach," he said. "This is a nice synergy between science and the computer -- the science pushing computer developments and the advanced computers pushing science forward, to the benefit of the science community and also the commercial world."

The calculations were performed under the U.S. Department of Energy's (DOE) Innovative and Novel Computational Impact on Theory and Experiment (INCITE) program on the Intrepid BlueGene/P supercomputer in ALCF at Argonne National Laboratory and on the Ds Cluster at Fermi National Laboratory, computer resources of the USQCD Collaboration. Part of the analysis was performed on the Iridis Cluster at the University of Southampton and the DiRAC facility in the UK. The research was supported by DOE's Office of Science, the U.K's Science and Technology Facilities Council, the University of Southampton, and the RIKEN Laboratory in Japan.

The Art of Rehearsing for the Courtroom

Somewhere along the continuum between the IBM Selectric and the iPad, courtroom presentations have moved from a formal debate podium to multi-media entertainment. Many courtrooms are wired like television studios and the medium may easily overshadow the message. As a result, rehearsal is more important than ever. Emerging, sophisticated technology has resulted in the ?never quite complete? presentation because trial teams have the ability to make last minute changes ?on the fly.? The advances in courtroom presentation technology have had an unintended consequence: attorneys fail to adequately rehearse the delivery of the presentation.

Few trial attorneys would dispute the notion that time spent rehearsing opening statements, mock trial presentations, and witness examinations is time well spent. But the most successful, experienced practitioners know that the actual time needed for effective, productive rehearsal sessions, is time that is difficult to schedule and time for which clients are loathe to pay. Additionally, rehearsal time is often sacrificed in the chaos of pre-trial activities such as jury instructions, in limine motions, client management, and settlement discussions. Rehearsing important presentations is a time-management issue and a trial priority. Excuses abound: ?I never rehearse, because I?m only good when I?m on,? or ?I don?t have enough time for this? or ?I?m best when I?m winging it? or ?I know this stuff cold; who needs to rehearse?? Rehearsals are about delivery of content so that delivery does not stand in the way of the message. Rehearsals in private, with colleagues, in front of a camera, and with presentation technology are the key to successful delivery of a winning style. This article outlines methods to effectively rehearse for effective courtroom presentations.

Rehearse is a verb

The Oxford American Dictionary defines rehearse: ?1. Practice (a play, recital, etc.) for later public performance. 2. Hold a rehearsal. 3. Train.? Rehearsal is about speaking out loud: essentially ?re-HEAR-ing.? Repetition and familiarity permit the brain to harmonize the key points throughout preparation of the presentation. Now that judges are imposing timing constraints at trial, rehearsals involve a stop watch component as well work on volume, clarity, pacing, and tone. Having participated in many previews, read-throughs, walk-throughs, run-throughs, practice sessions, dry-runs, and trial runs, there are reliably sound methods for effective rehearsal. The very fact that trial teams refer to this vital stage of trial preparation by so many different names is perhaps the clearest indication that the perception of its value is just as varied.

The past is prologue

In the late 80s, big poster boards, used to visually support an attorney?s remarks or witness testimony, were the result of developed black and white film onto which large sheets of color film were hand cut and applied. The boards were finalized, approved and assembled days before an opening to accommodate print production and assembly schedules. Similarly, slide carousel presentations were the result of designing content on photographic slide film, then cut and mounted within folding plastic frames, and then dropped into the carousel of the slide projector. These two state-of-the-art technologies (at the time) had the often overlooked advantage of creating the perfect scene for finalizing content while waiting for the slides or exhibit boards to be developed and delivered. After delivery of the slides and boards, trial teams rehearsed the delivery and coordination of the spoken word and visual content. Last minute changes on the first day of trial were not contemplated. Despite the wow factor of current technology, content still drives the heart of a presentation and that content is best served by a rehearsal process that polishes, tests, clarifies and perfects the spoken word with the medium.

Timing dictates rehearsal purpose

Each rehearsal should build upon the prior practice segment and feedback. The early ?recitals? are likely to be alone, in front of mirror, paper in hand, or even in the shower. The written scripts are the opportunity to hone the overall theme of the trial. Whether hand-written or digitally recorded, the early sessions are opportunities to improve upon the spoken content. One of the most common mistakes is that attorneys use the full script as the norm. It is exhausting to repeatedly rehearse a 60 or 90 minute presentation. Like actors in a play, attorneys are well advised to work on segments of the full presentation in short sessions with a defined purpose. Breaking the presentation into smaller thematic segments maintains a consistent level of performance and training. Also, rehearsing the transitions between segments is an often overlooked opportunity to refocus the audience on the overall theme of the case.

Current expectations for dynamic visuals have resulted in the need for another kind of rehearsal, the mechanical dry-run. It is where the lead attorney (not associates who guess what the ?boss? wants to see) choreographs the spoken and visual performance. The lead attorney is both director and actor during these ?choreographing? sessions; making decisions about which visuals to use, when to bring a visual up on the screen, and the transitions between thematic segments of the presentation. This process often results in reorganizing of key themes, thoughts and strategies for a more cohesive delivery.

Finally, full length rehearsals should be used sparingly. Rehearsal of the full presentation requires energy and focus. Experienced attorneys expect to spend up to 4 hours in a full dress rehearsal of a ninety-minute opening statement.

Assemble the right test-audience

After the content of an opening statement has essentially been written, edited, and vetted, it is ready for rehearsal sessions. Well-meaning associates might boost and soothe the ego, but often lack the experience to provide constructive feedback on presentation delivery. Ask experienced colleagues to attend and comment on the shorter practice sessions. Give each listener the permission to critique the performance and the content and ask for a written summary of the key points. Include non-lawyers, who also have no vested income in your feelings (i.e., avoid secretaries, parents, kids), in the audience.

Willingness to ask for, accept, and implement feedback is important throughout the process. For example, a lead trial attorney stays in the room for feedback during the early rehearsals, listening attentively and making on the spot improvements in her presentation. As the opening statement day draws nearer, she leaves the room after rehearsals to permit an unfiltered critique of her performance. Another senior trial attorney facilitates the feedback session, analyzing comments, and objectively using follow up questions to clarify the feedback on key points. This follow up analysis is then shared with the lead attorney in a private session that includes the trial consultant to achieve an effective and efficient result from the rehearsal.

Tackle the demon that is procrastination

When the BIG deadline is looming (opening statements for example), there?s nothing more counter-productive than failing to delegate tasks to the rest of the trial team. Most practitioners know to break large, intimidating tasks down to small, digestible bites to mark progress and complete the overall preparation of the case. Literally clearing a desktop, figuratively clears the mind. Reviewing trial textbooks or articles about the mechanics of oral presentations may break open a whole stream of consciousness that leads to an eloquent and persuasive delivery. Setting aside private time for reading the script out loud as opposed to doing other pretrial tasks creates space for final touch-ups.

There is often a reluctance to rehearse until everything is ?perfect,? and ?ready to go.? The main point of rehearsing at the early stages is to develop and refine presentation methodology. Waiting until the presentation is complete wastes good practice opportunities and causes unnecessary frustration and delay later in the trial preparation schedule. Many attorneys have wasted a rehearsal by shutting it down when the visuals weren?t perfect the first time. Remember the old adage, ?a picture is worth a thousand words?? In the courtroom, ?a picture represents a thousand words, but the attorney must choose those words wisely.?

Experienced trial attorneys make it look easy. Rest assured, those attorneys are the ones who rehearse the most, privately and publicly. ?Off the cuff,? is a romantic ideal, but is not a method for experienced trial attorneys. Attorneys who do not rehearse use the stress related adrenaline rush as a means of creating energy for last minute preparation. Feverishly finalizing content and supporting visuals throughout the night does not produce a persuasive performance. The best presentations hit the mark within two days of the opening. The lead attorney is able to ?rest? the night before and manage the ?peak? during the actual opening. A by-product of a timely final rehearsal is the luxury of editing. ?If I had more time, I?d write a shorter note,? applies here. Like it or not, the attention span of most juries are simply shorter and segmented. Teaching in 15-minute segments is most effective for any subject matter. Rehearsal of visuals, physical position and style of delivery all combine to punctuate and substantiate a compelling performance.

Though not a common problem among time-pressed attorneys, over-rehearsing deserves some mention. Rehearsing when tired or weary from subject matter familiarity produces a mechanical performance that lacks conviction and emotion. The audience is never engaged when the attorney is just ?going through the ?motions.??

The art of rehearsing is about training

Put simply, elite runners put in miles of training. Commitment to a training schedule requires purpose and drive. The discipline to use 5, 10, or 20 mile training runs is as important as diet, strength training, equipment and rest. Come race day, these runners, can trust the training and focus on peak performance. The same can be said for trial teams who apply this training discipline to the trial preparation schedule. Formal rehearsal schedules represent the final training regimen leading up to trial. Rehearsals are about speaking out loud with a focus on timing and delivery. Rehearse privately and with an audience. Rehearse in front of a camera and review the recording. Rehearse early and often with your trial consultant to review the demonstratives and exhibits timed to your script. Rehearse out loud with a test audience to hone the delivery. Well-rehearsed attorneys plant both feet firmly on the floor and confidently speak the tried and tested words that resonate with the jury in an easy and persuasive style.

Last Thursday we attended an event at the stunning Boodles store in the The Savoy Hotel, hosted by Brook-Dobson Brear. We were inspired by Michael Wainwright, Director of the British jewellery store previously known as ?Boodle and Dunthorne?.

Michael spoke about the transformation of their business and how they have reinvented their brand. We thought it was a great story of the journey of a business, very similar to the transformation our adviser clients have gone through in the lead up to RDR, so we?ve decided to share it with you on our blog.

Founded in 1798 in Liverpool, Boodle and Dunthorne became the city?s leading jewellers. After the sudden death of the Wainwright father and grandfather in 1945, sons Nicholas and Michael took over in 1992. Nicholas assumed the role of responsibility for creative direction of the company and Michael focused on developing the commercial side of the business.

With the growth of international jewellery brands, Michael and Nicholas knew they needed to become more competitive in such a tough market. Their vision was to become the British jewellery brand to compete head to head with the very best in the world. Driven by their desire to gain a more dominant position in the market, they started to look at developing their business and their brand. To drive such growth and achieve success they focused on a number of different elements within their business, and of course, their marketing.

Product

The original product strategy?was to buy in jewellery from a number of suppliers, including six very well-known watch brands. They realised that in order to differentiate themselves, they would need to achieve a more unique product offering. They took action and changed their strategy, hiring jewellery designers in-house in order to create more unique pieces and assign each a different name.

Boodles began focusing on the acquisition of rare and exquisite gemstones, as well as a highly talented in house design team, and the workmanship of leading British master craftsmen. Today Boodles offer over 400 different styles of jewellery, all from its eight main ranges including Raindance and Velocity.

Brand

The transformation of the brand began when the company was re-christened from ?Boodles and Dunthorne? to simply ?Boodles?. Dropping the second name they created a more simplistic, modern and luxurious brand image.

Next, Boodles began to focus on their image in order to differentiate the business from their competitors. They achieved this through looking at all elements of their image, and in particular, the photography used throughout their printed material and in store. Boodles began using their own models for photo shoots, shown wearing the jewellery outdoors, amongst beautiful scenery. This provided them with differentiation from other modern brands that elect to shoot in studio settings.

Client Experience

Aware of the critical importance of the client experience, Boodles decided to look at every element then reengineer the entire process. Focus was given to new shop fronts and internal displays. In collaboration with architect Eva Jiricna, the Boodles stores were transformed into gleaming confections of steel and glass ? the perfect backdrop for sparkling jewels and far removed from the ?country house? exposed beam interiors of the past.

Boodles also began to offer a service to all customers to upgrade their pieces of jewellery, every ten years to a newer product, thereby increasing the longevity of their customer relationships.

Community

To establish their brand image within the community and strengthen their customer relationships, Boodles decided to host and sponsor several large events, bringing together loyal clients and raising their corporate profile. These events include ?The Boodles Challenge Tennis Tournament?, ?Chester Races? and ?The Boodles Boxing Ball?.

Each year these events prove to be a great success, creating a sense of community around their brand. The fact that the events themselves are referred to by the name ?Boodles? is further testament to the strength of their brand.

Brand Expansion

To increase their market, Boodles decided to move their business international. Boodles had been considering a number of locations for international expansion, including the US and the Middle East, but have settled in Hong Kong after identifying Asia as a boom market. Boodles managing director Michael Wainwright said that he will look to mainland China for the company?s next Asian store, Beijing and Shanghai in particular. He also said that he hopes to open a Boodles store in New York within the next five years.

As a further expansion of their brand, Boodles have also decided to move their products online, creating a ?brick and clicks? retail business. Research carried out by Boodles in regards to their target customer, highlighted that they live quite busy lifestyles, with an online store creating added convenience for them.

By focusing on the key marketing elements of their business from product, to brand, to client experience, Boodles achieved and currently hold a dominant position in the jewellery market, competing head to head with the very best in the world.

We feel this is a very interesting and inspiring story that looks at a very effective marketing and business growth strategy. It is very similar to the work we do in our business, with financial advisers increasingly trying to transform their business and increase client engagement, in a changing regulatory environment.
